PEMANTAPAN MEDIA SUBKULTUR REGENERASI KALUS KEDELAI SECARA IN VITRO DENGAN PENAMBAHAN HORMON TUMBUH IBA DAN KINETIN

Abstract


This study aims to determine the in vitro enhancement of soy callus regeneration media with the addition of growth hormone Indole Butiric Acid (IBA) and kinetin. The research was conducted at the Laboratory of Bio-Science and Plant Reproduction Biotechnology, Department of Agricultural Cultivation, Faculty of Agriculture, Hasanuddin University (UNHAS), Makassar, from June 2020 to September 2020. This study used a completely randomized design (CRD) in a two-factor factorial pattern. The first factor, growth hormone IBA 5 levels: 0 ppm, 2.0 ppm, 4.0 ppm, 6.0 ppm, 8.0 ppm. The second factor, growth hormone Kinetin 4 levels: 0 ppm, 0.5 ppm, 1.0 ppm, 1.5 ppm. From these two factors obtained 20 treatment combinations. Each treatment was repeated 7 times, so that there were 140 experimental units. Data were analyzed using analysis of variance and LSD of 5%. The parameters observed were callus wet weight, callus color and callus structure. The results showed that Indole Butiric Acid (IBA) singly had a very significant effect on the increase in callus wet weight. The wet weight of callus was obtained at a concentration of 2.0 ppm IBA with an average of 0.81 grams, callus color with the highest scoring value was at a concentration of 6.0 ppm, namely 6.5 in the category between whitish green color and callus structure with the highest scoring value at a concentration of 8.0 ppm which is 2.8 in the category between compact callus structures there is a tendency to have a better effect. Kinetin alone had a very significant effect on the increase in callus wet weight. Wet weight time was obtained at 0.5 ppm kinetin with an average of 0.69 grams, callus color with the highest scoring value at a concentration of 1.5 ppm, namely 5.5 in the category between white and callus structure with the highest scoring value at concentration 1.0 ppm is 2.9 in the category between compact callus structures has a better effect. The combination of 2.0 ppm IBA growth regulators and 0.5 ppm kinetin resulted in a better callus wet weight of 0.89 grams. While the combination of IBA 6.0 ppm and kinetin 1.0 ppm, IBA 6.0 ppm and kinetin 1.5 ppm and IBA 8.0 ppm and kinetin 1.5 ppm resulted in better callus color (score score of 6.9 in the category between the whitish green color and the best callus structure in the combination treatment of IBA 6.0 ppm and kinetin 1.0 ppm with a score of 3.7 in the category of nodular compact callus structure.
